DltABCD-mediated d-alanylation of teichoic acids in Group A Streptococcus confers innate immune resistance

چکیده
Group A Streptococcus (GAS) is a major cause of both mucosal and invasive human infections. Epithelial and leukocyte production of cationic antimicrobial peptides (AMPs) is an important aspect of mammalian innate immune defense against bacterial infection. In this study, we identify a specific GAS phenotype that confers resistance to host AMPs. Inactivation of the dltA gene in an invasive serotype M1 GAS isolate led to loss of teichoic acid d-alanylation. Compared to the wild-type strain, the GAS dltA mutant was found to be more susceptible to AMP and lysozyme killing. Killing of the dltA mutant by human PMN, which produce AMPs in large amounts, was greatly accelerated. Thus, teichoic acid d-alanylation may contribute to the ability of invasive GAS to bypass mucosal defenses and produce systemic infection. D 2006 Published by Elsevier B.V.

D-alanylation of teichoic acids promotes group a streptococcus antimicrobial peptide resistance, neutrophil survival, and epithelial cell invasion.
Group A streptococcus (GAS) is a leading cause of severe, invasive human infections, including necrotizing fasciitis and toxic shock syndrome. An important element of the mammalian innate defense system against invasive bacterial infections such as GAS is the production of antimicrobial peptides (AMPs) such as cathelicidins. In this study, we identify a specific GAS phenotype that confers resis...
متن کاملD-Alanylation of Lipoteichoic Acids Confers Resistance to Cationic Peptides in Group B Streptococcus by Increasing the Cell Wall Density
Cationic antimicrobial peptides (CAMPs) serve as the first line of defense of the innate immune system against invading microbial pathogens. Gram-positive bacteria can resist CAMPs by modifying their anionic teichoic acids (TAs) with D-alanine, but the exact mechanism of resistance is not fully understood. Here, we utilized various functional and biophysical approaches to investigate the intera...
متن کاملEffect of D-alanylation of (lipo)teichoic acids of Staphylococcus aureus on host secretory phospholipase A2 action before and after phagocytosis by human neutrophils.
Invading bacteria such as Staphylococcus aureus induce mobilization of professional phagocytes (e.g., neutrophils) and extracellular antibacterial proteins (e.g., group IIA phospholipase A2 (gIIA PLA2)). Accumulation of gIIA PLA2 in inflammatory fluids confers potent extracellular antistaphylococcal activity and at lower concentrations promotes bacterial phospholipid degradation during phagocyt...
متن کاملVariations in the degree of D-Alanylation of teichoic acids in Lactococcus lactis alter resistance to cationic antimicrobials but have no effect on bacterial surface hydrophobicity and charge.
An increase of the degree of d-alanylation of teichoic acids in Lactococcus lactis resulted in a significant increase of bacterial resistance toward the cationic antimicrobials nisin and lysozyme, whereas the absence of D-alanylation led to a decreased resistance toward the same compounds. In contrast, the same variations of the D-alanylation degree did not modify bacterial cell surface charge ...

BACKGROUND Streptococcus gallolyticus subsp. gallolyticus (S. gallolyticus) is a pathogen of infective endocarditis. It was observed previously that this bacterium survives longer in macrophages than other species and the phagocytic uptake by and survival in THP-1 macrophages is strain-dependent. METHODS The phagocytosis assay was performed with THP-1 macrophages. S. gallolyticus specific who...
